Description

Sex difference

Aging is in large part depending on Vitamin D deficiency

Vit D synthesis requires heme

Heme synthesis is under estrogens control

Estrogens synthesis in peripheral tissues is dependent on DHEA supply from serum DHEAS.

Steroid sulfatase locally releases free DHEA from serum DHEAS.

Steroid sulfatase gene is on X chromosome, that usually undergoes inactivation to avoid excess genes expression in females.

Steroid sulfatase escapes X inactivation and therefore females have twice enzyme molecules than males. (steroid sulphatase and x inactivation)

During aging DHEAS levels are dropping and levels of steroid sulfatase may become the limiting factor for life extenction

Tumor necrosis factor inhibits conversion of dehydroepiandrosterone sulfate (DHEAS) to DHEA in rheumatoid arthritis synovial cells: a prerequisite for local androgen deficiency., 2005

These data indicate that TNF inhibits the conversion of DHEAS to DHEA in RA synovial cells. Because androgens are antiinflammatory mediators, TNF-induced inhibition of the local androgen supply is a supplementary proinflammatory factor.
