T-cell receptor (TCR) crosslinking leads to dephosphorylation of the tyrosine kinase LCK at Y505 by the membrane phosphatase CD45. Subsequent autophosphorylation of LCK at Y394 is an early event in T-cell activation, leading to phosphorylation (P) of downstream substrates by LCK. The tyrosine phosphatase PTPN22 and the intracellular kinase CSK both negatively regulate LCK by removal of the activating phosphate at Y394 (PTPN22) and rephosphorylation of Y505 (CSK). PTPN22 polymorphisms (R620W) have been shown to disrupt the binding of PTPN22 to CSK, and to increase the intrinsic phosphatase activity of PTPN22 itself.
